There will be 4 of us, all adults, going to WDW the last week of September and wanted to also go to Universal and IOA. Do we need a day for each park (Universal and IOA) or is it possible to cover both parks in one day?

As I think we would all say for Disney parks, at least a day per park to take your time, soak in the atmosphere, enjoy the small details that each ride and area have to offer.
while it is possible to do the major rides in each park in a single day (if you go commando style) you will miss a lot too. we have gone to universal several times over the past 3-4 years and we would never consider doing both parks in 1 day.

Yes, you should take a day for each park. Too much to see and do. Trying to do both in one day will make you rushed and you will miss too many attractions.
My advice and you can take it for what it is worth...
You can do both parks in 1 day. However, you should have a 1 night stay at one of their resorts. The reason I say this is their fastpass system is unlimited for people staying at their hotels. You also get into the park an hour early for Hogwarts (if they still do that).
If you purchase their fastpass thing, it is about $25-$45 per person per park. It is also only good for 1 ride on each of the rides included. You can get a room at Cabana for $139 on Sept 24.

To be clear, Cabana doesn't include the Universal Express - only deluxe resorts do. So depending on cost and number of people, it may be worth it to stay at a deluxe over a value - because of the pass.

Unless you go the route of booking a one night stay on site, definitely two days to do everything. If you are only going for WWoHP (both sides) that can be done in one. Make sure you get a park-to-park pass either way or you can't ride the train.

Like Disney parks, if your main goal is to run for rides only, one day each would probably work. If you want to slow down and see some of the shops and shows - take a day or two more. We were just there for 3 days and it was quite a pace and only did an overview. If your a HP fan, Diagon Alley can almost take a day in itself to see all details and shops.
