We will be staying at the DVC Vero beach resort just before our cruise, wondering if DCL transportation runs from VB resort? We have always just taken DCL transportation straight from MCO to port.
I've never seen any Disney cruise line buses at Disney's Vero beach. I think you will either need to rent a car or arrange for private transportation such as a town car service.

We just got back from Vero and I agree with Linda in that we have never seen any buses at the resort.
In my experience, DO NOT return a rental car at the port! The return at the cruise port is horrendous since it is no where near the terminal and you will have to take a shuttle from the rental car return. We have only had bad experiences with the shuttle buses since they only run infrequently and are on a first come first serve basis and will be very crowded.
Therefore, a private service seems to be your best option.
When we did a stay at Vero we had our own car that we parked at the cruise terminal.
